Interferon Gamma Elisa

Interferon Gamma ELISA is an immunoassay that measures interferon-gamma (IFN-γ), a cytokine released by activated immune cells, in biological samples. The method typically uses antibodies to capture IFN-γ and a second, enzyme-linked antibody to form a detectable antibody-antigen complex; adding a substrate produces a color change proportional to the cytokine concentration. In immunology and infection research, this assay quantifies cellular immune responses to antigens, supports evaluation of pathogen exposure and immune activation, and helps characterize inflammatory processes. Its sensitivity and quantitative readout make it useful for comparing experimental conditions, monitoring immune responses, and assessing candidate vaccines or immunotherapies.

Assay for the Identification of Interferon-Gamma Secretion in Murine Lymphoid Organs

This video demonstrates an imaging method to visualize in situ interferon-gamma (IFN-γ) secretion in the mouse spleen. Following bacterial infection and the induction of IFN-γ accumulation inside T cells and natural killer cells, the spleen is harvested from the mouse. Upon obtaining tissue sections, immunostaining is performed to visualize IFN-γ inside the cells.

Here, we describe a simple confocal imaging method to visualize the in situ localization of cells secreting the cytokine Interferon gamma in murine secondary lymphoid organs. This protocol can be extended for the visualization of other cytokines in diverse tissues.

The development of the interferon-gamma ELISpot assay for guinea pig PBMCs allows the characterization of T-cell responses in this highly relevant model for studying infectious diseases. We have applied the assay to measure T cell responses associated with intradermal delivery of DNA vaccines.
