Lc3-ii Cleavage

LC3-II cleavage is the enzymatic removal of lipid-conjugated LC3-II from autophagosomal membranes, a processing step that helps regulate autophagy. The cysteine protease ATG4 cleaves the amide bond linking LC3-II to phosphatidylethanolamine, releasing soluble LC3-I and enabling LC3 recycling; this differs from LC3 lipidation, which produces LC3-II from LC3-I. Measuring LC3-II cleavage alongside LC3 abundance, autophagosome formation, and lysosomal inhibition helps assess autophagic flux. In immunology and infection research, this analysis clarifies how immune cells control intracellular pathogens and how microbes alter autophagy-related pathways, supporting studies of host defense, inflammation, and therapeutic strategies.

Here, we present the methodology for concisely assessing autophagosome marker LC3-II levels in extracellular vesicles (EVs) by immunoblotting. This video demonstrates an assay to screen for the proteolytic activity of proteases using fluorogenic peptides. The protease recognizes its cleavage site on the peptide, cleaving it and separating the quencher from the fluorophore, enabling its fluorescence emission.
