Mucin Production Analysis

Mucin production analysis is the assessment of how epithelial cells synthesize and release mucins, heavily glycosylated proteins that form the mucus barrier protecting tissues from microbes and environmental damage. Researchers typically evaluate mucin abundance or secretion by measuring mucin gene expression, protein levels, or carbohydrate-rich material in cell cultures, tissue samples, or biological fluids, often after exposure to inflammatory signals or pathogens. In immunology and infection research, these measurements reveal how host cells modify barrier defenses, how microbial factors disrupt mucus production, and how immune signaling influences epithelial protection. The findings support studies of respiratory, gastrointestinal, and reproductive infections and can inform therapeutic strategies that restore mucosal function.

Using Unfixed, Frozen Tissues to Study Natural Mucin Distribution

Unfixed frozen tissue samples embedded in Optimal Cutting Temperature medium (OCT) can be used to study natural distribution and glycosylation of secreted mucus. In this approach tissue processing is minimal and the natural presentation of glycolipids, mucins and glycan-epitopes is preserved. Tissue sections can be analyzed by immunohistochemistry using fluorescence or chromogenic detection.

Mucin Agarose Gel Electrophoresis: Western Blotting for High-molecular-weight Glycoproteins

Mucins are high-molecular-weight glycoconjugates, with size ranging from 0.2 to 200 megadalton (MDa). As a result of their size, mucins do not penetrate conventional polyacrylamide gels and require larger pores for separation. We provide a detailed protocol for mucin agarose gel electrophoresis to assess relative quantification and study polymer assembly.

Measurement and Analysis of Extracellular Acid Production to Determine Glycolytic Rate

Glycolysis is a defining metabolic marker in multiple biological systems. Monitoring glycolysis by measuring the extracellular flux of H+ is common, but requires correction to be quantitative and unambiguous. Here, we demonstrate how to gather and correct extracellular flux data to distinguish between respiratory and glycolytic sources of extracellular acidification.
