Murine Cd11c+ Dendritic Cells

Murine CD11c+ dendritic cells are mouse immune cells identified by expression of the CD11c integrin and are widely studied as antigen-presenting cells that connect innate and adaptive immunity. In response to pathogens or inflammatory signals, they capture and process antigens, increase major histocompatibility complex class II and costimulatory molecules, and activate T cells through antigen presentation and cytokine production. Researchers use flow cytometry, cell isolation, genetic models, and infection experiments to examine dendritic-cell development, trafficking, and function. These studies clarify how immune responses are initiated and regulated, supporting investigation of host defense, inflammation, vaccination, and infectious disease mechanisms.

In Vitro Generation of Plasmacytoid Dendritic Cells from Common Lymphoid Progenitors

This video demonstrates an in vitro assay to generate plasmacytoid dendritic cells (pDCs) from common lymphoid progenitor cells (CLPs). Upon co-culturing purified CLPs with γ-irradiated feeder cells in the presence of Fms-like tyrosine kinase-3 ligand (FL), the CLPs differentiate into non-adherent pDCs, the presence of which is confirmed using flow cytometry.

There is an increasing interest in understanding the immunological functions of specific subpopulations of cells in Peyer's patches (PPs), the primary inductive sites of gut-associated lymphoid tissues. Here we outline parallel protocols for preparing PP single cell preparations for flow cytometric analysis and PP cryosections for immunostaining.
