Parasite Burden

Parasite burden is the quantity of parasites present in a host, tissue, or population, and it is a key measure of infection severity and transmission potential. It reflects the balance between parasite replication and host clearance, shaped by exposure dose, tissue distribution, immune responses, and treatment. In immunology and infection research, investigators estimate parasite burden using microscopy, culture, molecular detection, or organism counts to compare disease progression and treatment efficacy. Tracking changes over time helps link pathogen load with inflammation, immune dysfunction, clinical outcomes, and the development of interventions that reduce infection and limit transmission.

An in vivo imaging system is used to generate quantitative measurements of murine infection with the Trypanosomatid protozoan Leishmania. This is a non-invasive and non-lethal method for detecting parasites expressing luciferase within many tissues throughout the course of chronic Leishmania spp. infection.

Heligmosomoides polygyrus is a murine nematode with powerful immunomodulatory capabilities that closely resemble those of highly-prevalent human helminth infection. Here we describe a protocol for the long-term maintenance of the H. polygyrus lifecycle.

Assessing Bacterial Burden in the Mouse Spleen for Infection Studies

Source: Madenspacher, J. H. and Fessler, M. B. A Non-invasive and Technically Non-intensive Method for Induction and Phenotyping of Experimental Bacterial Pneumonia in Mice. J. Vis. Exp. (2016)This video demonstrates the process of quantifying bacterial load in the spleen of an infected mouse to assess the severity of infection. The homogenized spleen is serially diluted and plated to determine total bacterial burden. The resulting data provide insights into host-pathogen interactions within...

Protocol for Production of a Genetic Cross of the Rodent Malaria Parasites

Genetic crosses of rodent malaria parasites are performed by feeding two genetically distinct parasites to mosquitoes. Recombinant progeny are cloned from mouse blood after allowing mosquitoes to bite infected mice. This video shows how to produce genetic crosses of Plasmodium yoelii and is applicable to other rodent malaria parasites.

An In Vitro Assay to Quantify the Intracellular Growth of Parasite in Macrophages

2025

This video demonstrates an assay to quantify the intracellular growth of parasites in bone marrow-derived macrophages (BMMs). The macrophage cells are infected with the motile and non-motile invasive stages of Leishmania, metacyclic promastigotes and amastigotes, respectively. The parasites invade the macrophages, getting internalized into a parasitophorous vacuole and multiplying inside. The intracellular parasite growth is quantified using a microscope upon fluorescently staining the...
