2d Planar Imaging

2D planar imaging is a medical imaging method that produces two-dimensional projection views of anatomical structures or physiological activity, supporting diagnosis and treatment planning. In nuclear medicine, a patient receives a radiotracer that emits gamma photons; a collimator directs the photons toward a gamma camera, where a scintillation crystal converts them into light and electronic signals to form an image. Planar imaging is used to assess radiotracer distribution in applications such as bone, thyroid, renal, and cardiac studies. Although it provides less depth information than tomographic techniques, its speed, broad availability, and ability to show whole-body tracer patterns make it valuable in clinical evaluation.

This method estimates the absorbed dose of different structures for peptide-receptor-radionuclide-therapy (PRRT) with the possibility of avoiding organ overlap on 2D-projections. Serial whole-body planar images permit estimation of mean absorbed doses along the whole body, while the hybrid approach, combining planar images and 3D-SPECT/CT image, overcomes the limitations of structure overlapping.

Adaptive immunity is controlled by dynamic 'immunological synapses' formed between T cells and antigen presenting cells. This protocol describes methods for investigating endothelial cells both as understudied physiologic APCs and as a novel type of 'planar cellular APC model'.

A scanner for imaging magnetic particles in planar samples was developed using the planar frequency mixing magnetic detection technique. The magnetic intermodulation product response from the nonlinear nonhysteretic magnetization of the particles is recorded upon a two-frequency excitation. It can be used to take 2D images of thin biological samples.

We describe here a combination of the glass-supported lipid bilayer technique of forming immunological synapses with the super-resolution imaging technique of stimulated emission depletion (STED) microscopy. The goal of this protocol is to provide users with the instructions necessary to successfully carry out these two techniques.
