Human Retinal Specimens

Human retinal specimens are samples of retinal tissue used to examine the structure, cell types, and molecular features of the light-sensitive neural tissue at the back of the eye. Their scientific value depends on preserving the retina’s layered architecture and cellular components so researchers can apply histology, immunohistochemistry, microscopy, and molecular analyses to identify disease-related changes. In medicine, these specimens support investigation of conditions such as diabetic retinopathy, age-related macular degeneration, glaucoma, and inherited retinal disorders. Findings can clarify disease mechanisms, reveal potential biomarkers, and help evaluate whether experimental treatments target the relevant human tissues.

Transcriptomic Analysis of Human Retinal Surgical Specimens Using jouRNAl

We used retinal samples from retinectomy for a transcriptomic analysis of retinal detachment. We developed a procedure that allows RNA conservation between the surgical blocks and the laboratory. We standardized a protocol to purify RNA by cesium chloride ultracentrifugation to assure that the purified RNAs are suitable for microarray analysis.

Here, a protocol to isolate and establish primary fibroblast/myofibroblast (MF) cultures from frozen gastric, small intestinal, and colonic tissue-yielding cells with a MF phenotype-is presented. These cells express CD90, α-SMA and vimentin. MFs can be used for a variety of functional assays including enzymatic activity and cytokine production.

Improved Protocol For Laser Microdissection Of Human Pancreatic Islets From Surgical Specimens

Laser microdissection is a technique that allows the recovery of selected cells from minute amounts of parenchyma. Here we describe a protocol for acquiring human pancreatic islets from surgical specimens to be used for transcriptomic studies. Our protocol improves the intrinsic autofluorescence of human beta cells, thus facilitating their collection.
