Tumor Stroma Analysis

Tumor stroma analysis is the systematic study of the nonmalignant tissues, cells, and extracellular matrix that surround and interact with cancer cells. By combining histology, immunohistochemistry, molecular profiling, or imaging, researchers characterize components such as fibroblasts, immune cells, blood vessels, and matrix proteins, then assess how their signals and physical properties influence tumor growth, invasion, immune evasion, and treatment response. In medicine, this analysis helps define tumor subtypes, identify prognostic and predictive biomarkers, clarify mechanisms of therapeutic resistance, and guide development of approaches that target both malignant cells and their surrounding microenvironment.

A Novel Stromal Fibroblast-Modulated 3D Tumor Spheroid Model for Studying Tumor-Stroma Interaction and Drug Discovery

A novel three-dimensional spheroid model based on the heterotypic interaction of tumor cells and stromal fibroblasts is established. Here, we present coculture of tumor cells and stromal fibroblasts, time-lapse imaging, and confocal microscopy to visualize the formation of spheroids. This three-dimensional model offers a pertinent platform to study tumor-stroma interactions and test cancer therapeutics.

Luciferase-modified human brain tumor xenografts can be established intracranially in athymic mice, with subsequent monitoring of tumor growth and response to therapy using bioluminescence imaging. In combination with survival analysis, bioluminescence monitoring is an essential research tool for pre-clinical testing of therapies being considered for treating brain tumors.
