Intestinal Ligated Loop

The intestinal ligated loop is an in vivo experimental technique that isolates a defined segment of the intestine to investigate local physiological and pathological responses. In this method, surgeons place ligatures around selected intestinal sections and introduce a test substance, microorganism, or formulation into the enclosed loop, allowing changes in fluid movement, absorption, permeability, or tissue response to be measured under controlled conditions. In medicine and pharmacology, the model supports studies of intestinal transport, drug and vaccine delivery, inflammation, and host-pathogen interactions. By linking localized exposure to measurable tissue and luminal outcomes, it provides valuable evidence for gastrointestinal research and therapeutic development.

M cells in a specialized follicle-associated epithelium covering Peyer’s patches play an important role for the mucosal immunosurveillance in gut-associated lymphoid tissue. Here we described the evaluation method for bacterial transcytosis by M cells in vivo. This method provides a method to understand M-cell function in the immune system.

We describe a novel surgical method for catheterizing 'intestinal loops' within the ileum of sheep. Once animals have recovered from surgery and have cleared antibiotics and analgesics, multiple treatments can be deposited directly in loops via the catheters.

Here we present a protocol to surgically create 'intestinal ligated loops' in chicken small intestines. This procedure allows for the comparison of multiple Clostridium perfringens strains' virulence in situ in a single host. This method markedly decreases the number of chickens usually necessary for similar in vivo experiments.
