Lipid-laden Hepatocytes

Lipid-laden hepatocytes are liver cells containing excessive intracellular lipid droplets, a hallmark of hepatic steatosis and an important indicator of metabolic or toxic injury. They arise when fatty acid uptake and synthesis exceed mitochondrial oxidation and triglyceride export, causing lipids to accumulate within the cytoplasm and potentially disrupting hepatocyte function. In medicine, identifying these cells through histopathology, imaging, or laboratory models supports evaluation of fatty liver disease, metabolic dysfunction, and drug-induced liver injury. Their abundance and morphology can help researchers assess disease progression, treatment responses, and how candidate therapies alter hepatic lipid metabolism.

Oil Red O Staining: A Technique for Staining Neutral Lipids in Hepatocytes for Detecting Hepatic Steatosis In Vitro

In this video, we demonstrate in vitro staining of neutral lipids in hepatocytes to detect hepatic steatosis or excessive fat accumulation in liver. Excessive fat is accumulated in the cells in form of lipid droplets. Oil Red O is a lipid soluble dye that binds to intracellular neutral lipids in these lipid droplets. When observed under a light microscope, these intracellular lipid droplets appear intensely red in color.

Isolation of Human Hepatocytes by a Two-step Collagenase Perfusion Procedure

A modified two-step collagenase perfusion procedure for isolation of human hepatocytes is described. This method can also be applied to other mammalian livers. The isolated hepatocytes are available in high yield and viability, making them a suitable model for scientific research in areas such as liver regeneration, pharmacokinetics and toxicology.

Here, we show an enzymatic approach to isolate primary hepatocytes from adult mice, and we describe the quantification of an inflammatory response using ELISA and real-time PCR.

Preparation and Delivery of Protein Microcrystals in Lipidic Cubic Phase for Serial Femtosecond Crystallography

We describe procedures for the preparation and delivery of membrane protein microcrystals in lipidic cubic phase for serial crystallography at X-ray free-electron lasers and synchrotron sources. These protocols can also be applied for incorporation and delivery of soluble protein microcrystals, leading to substantially reduced sample consumption compared to liquid injection.
