Retinal Pathophysiology

Retinal pathophysiology is the study of how disease, injury, or abnormal development disrupts the retina’s structure and function, impairing visual processing. These changes can affect photoreceptors, retinal neurons, blood vessels, or supporting cells, altering light detection, cellular signaling, and transmission of information to the optic nerve. Understanding these mechanisms helps explain vision loss in conditions such as diabetic retinopathy, age-related macular degeneration, retinal detachment, and inherited retinal disorders. Retinal pathophysiology supports clinical diagnosis, interpretation of imaging and functional tests, development of targeted treatments, and research into strategies for preserving or restoring vision.

We provide a reproducible method for culturing confluent monolayers of human fetal retinal pigment epithelial cells (hfRPE) cells that exhibit morphology, physiology, polarity, and protein and gene expression patterns of adult native tissue. This work has been extended to an animal model of several eye diseases.

Diabetic retinopathy is one of the leading causes of blindness. Histology, blood-retinal barrier breakdown assay, and fluorescence angiography are valuable techniques to understand the pathophysiology of the retina, which could further enhance the efficient drug screening against diabetic retinopathy.

Pathophysiology of Diabetes

Diabetes mellitus is a chronic metabolic disorder characterized by hyperglycemia. The four categories of diabetes are type 1 diabetes, type 2 diabetes, other specific types of diabetes, and gestational diabetes. Type 1 diabetes is characterized by autoimmune-mediated destruction of pancreatic β cells, with environmental factors potentially triggering this process in genetically susceptible individuals. Despite many not having a family history, certain genes increase susceptibility, suggesting a...
