Hippocampal Theta Oscillations

Hippocampal theta oscillations are rhythmic fluctuations in electrical activity, typically occurring at about 4–12 Hz, that organize neural processing in the hippocampus and support memory-related functions. They arise through coordinated interactions among hippocampal circuits, including excitatory pyramidal cells and inhibitory interneurons, and their timing can align neuronal firing to distinct phases of the theta cycle. In neuroscience, researchers study theta power, phase, and relationships with other brain regions to examine navigation, learning, memory formation, and sleep-wake states. Measuring these oscillations with electrophysiology or imaging helps reveal how brain networks coordinate information and may clarify circuit dysfunction in neurological disease.

We describe the use of optogenetics and electrophysiological recordings for selective manipulations of hippocampal theta oscillations (5-10 Hz) in behaving mice. The efficacy of the rhythm entrainment is monitored using local field potential. A combination of opto- and pharmacogenetic inhibition addresses the efferent readout of hippocampal synchronization.

Here, we present a protocol for recording rhythmic neuronal network theta and gamma oscillations from an isolated whole hippocampal preparation. We describe the experimental steps from extraction of the hippocampus to details of field, unitary and whole-cell patch clamp recordings as well as optogenetic pacing of the theta rhythm.
