Microglia Biomarkers

Microglia biomarkers are measurable molecular, cellular, or imaging features used to identify microglial presence, abundance, phenotype, or activity in the nervous system. They work by detecting microglia-associated proteins or transcripts, such as IBA1, TMEM119, and P2RY12, or by measuring targets such as TSPO with imaging; interpretation depends on tissue context and assay specificity because no single marker captures every microglial state. In neuroscience, these biomarkers support immunohistochemistry, flow cytometry, transcriptomic profiling, and positron emission tomography, helping researchers compare homeostatic and disease-associated microglia in conditions including neurodegeneration, inflammation, injury, and brain development.

We describe a rat model of post traumatic stress disorder (PTSD) that reveals the persistent alterations in neuroendocrine function and the delayed long-term, exaggerated fear response, characteristic of PTSD patients. The animal model and methods described here are useful for correlating biomarkers in brain nuclei, which are mechanistic but cannot be measured in patients, with biomarkers in peripheral white blood cells, which can.

Magnetic Isolation of Microglia from Mouse Pup Brains

In this video, we describe a protocol for isolating CD11b cells, including microglia, from mouse pup brains using magnetic-activated cell sorting. The brain tissues are homogenized to obtain single-cell suspensions, which are incubated with anti-CD11b antibody-coated microbeads, followed by a magnetic column-based separation to purify the CD11b+ cell population.

Culturing Microglia from the Neonatal and Adult Central Nervous System

We outline methods for the efficient and quick isolation/culture of viable microglia from the neonatal cerebral cortex and adult spinal cord. The dissection and plating of cortical microglia can be accomplished within 90 minutes, with the subsequent microglial harvest taking place ~ 10 days following the initial dissection.

Microglia (immune cells of the brain), are used as a surrogate biosensor to determine how nanoparticles influence neurotoxicity. We describe a series of experiments designed to assay microglial response to nanoparticles and exposure of hypothalamic neurons to supernatant from activated microglia to determine neurotoxicity.
