Mounting Medium Preservation

Mounting medium preservation is the use of a protective medium to stabilize microscope specimens between a slide and coverslip, maintaining their structure and optical quality during imaging and storage. The medium surrounds the tissue, limits dehydration and physical disturbance, and can improve light transmission by matching the refractive index of the specimen and coverslip; some formulations also help preserve fluorescent labels. In neuroscience, it is applied to stained brain sections and neuronal preparations to retain cellular morphology, axonal projections, and anatomical signals for fluorescence or bright-field microscopy. Consistent mounting supports reliable image comparison, archiving, and quantitative analysis.

A Method for Generating Oligodendrocyte-Conditioned Medium

2025

This video demonstrates the production of oligodendrocyte-conditioned medium (OCM) from oligodendrocyte lineage cells by stressing them to enhance their molecular release into the medium. This medium, which is rich in growth factors, cytokines, and other bioactive molecules, can be added to neuron cultures to gain insight into the impact of oligodendrocytes-secreted factors on neuronal physiology and connectivity.

Whole-Mount In Situ Hybridization

2023

Whole-mount in situ hybridization (WMISH) is a common technique used for visualizing the location of expressed RNAs in embryos. In this process, synthetically produced RNA probes are first complementarily bound, or "hybridized," to the transcripts of target genes. Immunohistochemistry or fluorescence is then used to detect these RNA hybrids, revealing spatial and temporal patterns of gene expression. Unlike traditional in situ hybridization techniques, which require thin tissue sections whose...
