Neuronal Glial Co-culture

Neuronal glial co-culture is an in vitro model that grows neurons together with glial cells to reproduce key cellular interactions within nervous tissue. In this system, glia support neuronal survival and maturation while regulating the extracellular environment, providing trophic signals, and influencing synaptic communication through direct contact and secreted factors. Researchers use co-culture models to study neurodevelopment, synapse formation, neural signaling, inflammation, and responses to injury or disease. By incorporating multiple neural cell types rather than neurons alone, neuronal glial co-culture can provide more physiologically relevant insights for neuroscience research and evaluation of potential therapeutics.

Obtaining a Mixed Glial Cell Culture from a Neonatal Mouse Brain

2025

The video demonstrated the method of isolating and culturing cells from a neonatal mouse brain. It involved enzymatic and mechanical dissociation of the brain, followed by culturing in growth media lacking neuronal growth factor to obtain a mixed glial population.

Establishing a Müller Glial Cell Culture Obtained from Mouse Retinas

2025

This video demonstrates the culturing of Müller glial (MG) cells isolated from mouse retinas. Retinas are dissociated into a single-cell suspension using digestive enzymes. Cells are then grown in a medium with a growth factor that promotes MG cell proliferation and neuronal cell mortality. Dead neuronal cells are removed while passaging, and MG cells are grown for further analysis.

Primary Neuronal Cultures

2023

The complexity of the brain often requires neuroscientists to use a simpler system for experimental manipulations and observations. One powerful approach is to generate a primary culture by dissecting nervous system tissue, dissociating it into single cells, and growing those cells in vitro. Primary cultures make neurons and glia easily accessible to the experimental tools required for techniques like genetic manipulation and time-lapse imaging. Furthermore, these cultures represent a highly...

A Technique to Culture Mouse Hippocampal Neurons

2025

The video demonstrates a method for culturing neurons from murine hippocampi. Initially, the hippocampi undergo enzymatic digestion to break down the extracellular tissue matrix. Subsequently, the digested tissue is mechanically disrupted to isolate individual neurons. Finally, these neurons are cultured on glial feeder cells.

This article describes the protocol for culturing low-density primary hippocampal neurons growing on glass coverslips inverted over a glial monolayer. The neuron and glial layers are separated by paraffin wax beads. The neurons grown by this method are suitable for high-resolution optical imaging and functional assays.
