Active Galactic Nuclei

Active Galactic Nuclei (AGN) are exceptionally bright, compact regions at the centers of some galaxies, powered by material falling toward a supermassive black hole. As gas and dust form a rapidly rotating accretion disk, gravitational energy is converted into intense radiation, while magnetic fields can help launch collimated jets and powerful outflows. AGN emit across the electromagnetic spectrum, from radio waves to X-rays, making them valuable laboratories for studying black hole growth, plasma physics, and relativistic processes. Their radiation and outflows can also influence star formation and the evolution of their host galaxies, linking small-scale accretion to large-scale cosmic structure.

This video describes the protocol to isolate single-nuclei from fresh frozen glioma tissue. The isolated single-nuclei can be used as specimens for sequencing techniques such as single-nuclei RNA (snRNA) sequencing and single-nuclei Assay for Transposase-Accessible Chromatin (snATAC) sequencing to obtain deeper insights into inter- and intra-tumor heterogeneity observed in glioma of the central nervous tissues.

Presented here is a method to sequence single nuclei isolated from the mouse dentate gyrus that excludes most neurons through fluorescence-activated nuclei (FAN)-sorting. This approach generates high-quality expression profiles and facilitates the study of most other cell types represented in the niche, including scarce populations such as neural stem cells.
