Single Cell Flow System

A Single Cell Flow System is a fluid-handling platform that transports, isolates, and analyzes individual cells, enabling biochemical measurements that would be obscured in bulk samples. It typically uses microfluidic channels and controlled pressure or flow to guide cells through defined regions, where optical, electrical, or chemical signals can be recorded one cell at a time. By linking cell position and behavior to measurable molecular properties, the system supports investigations of cellular heterogeneity, enzyme activity, signaling, and responses to biochemical conditions. These measurements can reveal rare cell states, improve assay sensitivity, and advance research in diagnostics, drug development, and systems biology.

Microbial biofilms are generally constituted by distinct subpopulations of specialized cells. Single-cell analysis of these subpopulations requires the use of fluorescent reporters. Here we describe a protocol to visualize and monitor several subpopulationswithin B. subtilis biofilms using fluorescence microscopy and flow cytometry.

An efficient method to obtain highly purified viable meiotic fractions from mouse testis is described, which combines a refined cell dissociation protocol with fluorescent activated cell sorting (FACS). This method takes advantage of differences in the DNA content and nuclear density of discrete meiotic fractions.

Intracellular Ca2+ dynamics are very important in sperm physiology and Ca2+-sensitive fluorescent dyes constitute a versatile tool to study them. Population experiments (fluorometry and stopped flow fluorometry) and single cell experiments (flow cytometry and single cell imaging) are used to track spatio-temporal [Ca2+] changes in human sperm cells.

Visualizing Cytoplasmic Flow During Single-cell Wound Healing in Stentor coeruleus

The giant ciliate Stentor coeruleus is a classical system for studying regeneration and wound healing in single cells. By imaging Stentor cells simultaneously at low and high magnification it is possible to measure cytoplasmic flows before, during, and after wounding.
