Sumo-trapping Protein

A SUMO-trapping protein is a molecular reagent that selectively captures SUMO-modified proteins, enabling analysis of this reversible post-translational modification in biochemistry. Typically, it uses the substrate-recognition surface of a SUMO protease or a SUMO-interacting motif while lacking catalytic activity, allowing it to bind SUMO conjugates without cleaving the isopeptide bond that links SUMO to a target protein. Researchers use these traps to enrich SUMOylated proteins from cell or tissue extracts for immunoblotting, purification, and mass spectrometry. By stabilizing otherwise transient interactions, the approach helps characterize SUMO-regulated pathways, identify modification targets, and investigate cellular responses to stress, DNA damage, and other signals.

SUMO is an essential and highly conserved, small ubiquitin-like modifier protein. In this protocol we are describing the use of a stress-tolerant recombinant SUMO-trapping protein (kmUTAG) to visualize native, untagged SUMO conjugates and their localization in a variety of cell types.

This protocol describes the method of gene trap insertional mutagenesis using Gal4-VP16 as the primary reporter and GFP/RFP as secondary reporters in zebrafish. Approximately one in ten high-expressing F0 fish yield gene trap progeny co-expressing GFP and RFP. The screening procedure can be readily scaled to adapt to the size of the laboratory performing the insertional mutagenesis screen.
