White Blood Cell Cytometry

White blood cell cytometry is a quantitative method for measuring and characterizing leukocytes, providing information about their abundance, size, internal complexity, and molecular identity. In flow cytometric systems, cells pass individually through a focused fluid stream, where laser-induced light scatter and fluorescence from labeled antibodies are detected and converted into cellular profiles. These measurements enable immunophenotyping, cell counting, and identification of distinct immune cell populations. In bioengineering, white blood cell cytometry supports immune monitoring, biomaterial and drug evaluation, cell-based product development, and the design of diagnostic systems by linking cellular responses to engineered environments or therapeutic interventions.

This video demonstrates a flow cytometry-based technique using two fluorochromes to identify CD4+ and CD8+ T cells, γδ T cells, B cells, natural killer (NK) cells, and monocytes in human peripheral blood mononuclear cells (PBMCs). Upon incubating the cells with fluorochrome-conjugated antibodies against the surface markers, the cell subsets are identified using an optimal flow cytometric gating strategy.

An efficient method to obtain highly purified viable meiotic fractions from mouse testis is described, which combines a refined cell dissociation protocol with fluorescent activated cell sorting (FACS). This method takes advantage of differences in the DNA content and nuclear density of discrete meiotic fractions.

This paper presents a flow cytometry-based method to investigate the immune composition of aortas. The paper also illustrates an additional technique that allows examining surrounding adventitia and vessel wall separately. This method opens possibilities to perform phenotypical analyses of aortic leukocytes and apply several immunological assays for atherosclerosis studies.
