Cell Culture Optimization

Cell culture optimization is the systematic adjustment of laboratory conditions to maintain healthy, reproducible cell growth and function, which is essential for reliable biological research. It works by balancing factors such as culture medium composition, temperature, pH, osmolality, seeding density, and passage timing while monitoring cell viability, morphology, and proliferation. Optimized cultures support experiments in developmental biology, disease modeling, drug screening, tissue engineering, and biotechnology by reducing variability and improving experimental consistency. Careful optimization can also extend culture performance, preserve cell-specific characteristics, and strengthen the reproducibility of results across researchers and laboratories.

Protocol for propagation of dissociated high grade glioma surgical specimens in serum-free neurosphere medium to select for cells with cancer stem cell phenotype. For specimens that fail to grow as neurospheres, an alternative protocol is suggested. A paraffin embedding technique for maintaining the 3D neurosphere architecture for immunocytochemistry is described.

This manuscript outlines a detailed video protocol for culturing primary lens epithelial cells (LECs), aiming to improve reproducibility and aid research in cataracts and posterior capsule opacification (PCO). It offers step-by-step instructions on lens dissection, LECs isolation, and validation, serving as a valuable guide, especially for newcomers in the field.

Rotating Cell Culture Systems for Human Cell Culture: Human Trophoblast Cells as a Model

Traditional, two dimensional cell culture techniques often result in altered characteristics with respect to differentiation markers, cytokines and growth factors. Three-dimensional cell culture in the rotating cell culture system (RCCS) reestablishes expression of many of these factors as shown here with an extravillous trophoblast cell line.

Here is a protocol to grow pluripotent stem cells (PSC) and neural stem cells (NSC) in an enclosed cell culture system that permits maximum sterility and reproducibility, replacing the traditional biosafety cabinet and incubator. This equipment meets clinical good manufacturing practice (cGMP) and clinical good lab practice (cGLP) guidelines.
