Dominant Follicle Selection

Dominant follicle selection is the reproductive process in which one ovarian follicle gains developmental priority and becomes the follicle most likely to ovulate. During the follicular phase, a cohort of follicles responds to follicle-stimulating hormone (FSH), but the follicle with greater FSH sensitivity continues growing as FSH levels decline; its production of estradiol and inhibin further suppresses FSH, promoting atresia of competing follicles. This process coordinates ovulation, regulates the number of oocytes released, and supports reproductive timing. Studying dominant follicle selection helps explain ovarian physiology, menstrual-cycle regulation, fertility, multiple ovulation, and responses to assisted reproductive treatments.

The Utility of Stage-specific Mid-to-late Drosophila Follicle Isolation

Stage-specific isolation of mid-to-late Drosophila follicles is useful for a variety of purposes. Such follicles develop in culture, which allows for genetic and/or pharmacologic manipulations to be coupled with in vitro development assays and live imaging. Additionally, follicles can be used for molecular studies, such as isolating mRNA and protein.

This protocol describes the primary culture/co-culture of mouse ovarian tissue, using ovaries from neonatal mice and individual ovarian follicles from prepubertal mice. The culture techniques support development in a highly physiological manner, allowing investigation of the effect of extrinsic agents on the ovary, and of interactions between ovarian follicles.
