Kidney Gene Therapy

Kidney gene therapy is an approach that treats renal disease by delivering genetic material to kidney cells, with the goal of correcting harmful mutations or changing disease-related gene activity. It can use viral or nonviral vectors to transport a functional gene, gene-editing components, or regulatory sequences into target cells, where they may restore protein production or modify abnormal signaling. In biology research, kidney gene therapy supports investigation of inherited disorders, chronic kidney injury, and mechanisms of renal repair. Successful development could reduce dependence on dialysis or transplantation while advancing precision treatments, though efficient delivery and long-term safety remain important challenges.

Gene Therapy

2021

Gene therapy is a technique where a gene is inserted into a person’s cells to prevent or treat a serious disease. The added gene may be a healthy version of the gene that is mutated in the patient, or it could be a different gene that inactivates or compensates for the patient’s disease-causing gene. For example, in patients with severe combined immunodeficiency (SCID) due to a mutation in the gene for the enzyme adenosine deaminase, a functioning version of the gene can be inserted. The...

In this protocol, baculovirus is produced by transient transfection of baculovirus plasmid into Sf9 cells and amplified in a serum-free suspension culture. The supernatant is purified by heparin affinity chromatography and further concentrated by ultracentrifugation. This protocol is useful for the production and purification of baculovirus for gene therapy application.

In this report we describe a method for the isolation and culture of the progenitor cell niche from the embryonic mouse kidney that can be used to study signaling pathways regulating stem/progenitor cells of the developing kidney. These cultured cells are highly accessible to small molecule and recombinant protein treatment, and importantly also to viral transduction, which allows efficient manipulation of candidate pathways.
