Orf Expression Library

An ORF expression library is a collection of cloned open reading frames (ORFs), each representing a protein-coding sequence, assembled for controlled gene expression and functional analysis. In these libraries, individual ORFs are inserted into expression vectors and placed under regulatory elements such as promoters; after introduction into a suitable host, the host’s transcription and translation machinery produces the encoded proteins. Researchers use ORF expression libraries to test gene function, compare protein activity, identify interactions, and screen candidate targets in biology and biotechnology. By enabling systematic analysis of many genes in parallel, they support functional genomics, pathway studies, and development of protein-based research tools.

Construction of a fosmid library with environmental genomic DNA isolated from the vertical depth continuum of a seasonally hypoxic fjord is described. The resulting clone library is picked into 384-well plates and archived for downstream sequencing and functional screening by the application of an automated colony picking system.
