Transforming Growth Factor Beta

Transforming Growth Factor Beta (TGF-β) is a family of multifunctional cytokines that regulates cell growth, differentiation, tissue maintenance, and immune responses. Signaling begins when TGF-β binds type II and type I serine/threonine kinase receptors, triggering phosphorylation of SMAD proteins that form complexes, enter the nucleus, and alter gene transcription; additional non-SMAD pathways can also contribute. In biology, TGF-β signaling helps coordinate embryonic development, wound healing, extracellular matrix production, and immune regulation. Dysregulated activity is associated with fibrosis, inflammatory disease, and cancer, making the pathway an important focus for understanding disease mechanisms and developing targeted therapies.

A protocol for in vitro induction of endothelial-mesenchymal transition (EndMT), which is useful for investigating cellular signaling pathways involved in EndMT, is described. In this experimental model, EndMT is induced by treatment with TGF-β in MS-1 endothelial cells.

We have modified the conventional yeast two-hybrid screening, an effective genetic tool in identifying protein interaction. This modification markedly shortens the process, reduces the workload, and most importantly, reduces the number of false positives. In addition, this approach is reproducible and reliable.

A Simple Bioassay for the Evaluation of Vascular Endothelial Growth Factors

We describe a simple cell-based bioassay for detecting, quantifying and monitoring the activity of members of the vascular endothelial growth factor family of ligands. The assay uses chimeric receptors expressed in a factor-dependent cell line to provide a semi-quantitative or quantitative assessment of receptor binding and cross-linking by the ligand.

This protocol combines electrospinning and microspheres to develop tissue engineered scaffolds to direct neurons. Nerve growth factor was encapsulated within PLGA microspheres and electrospun into Hyaluronic Acid (HA) fibrous scaffolds. The protein bioactivity was tested by seeding the scaffolds with primary chick Dorsal Root Ganglia and culturing for 4-6 days.
