Umbilical Arteries

Umbilical arteries are paired fetal blood vessels that run through the umbilical cord and connect fetal circulation with the placenta. They carry relatively deoxygenated blood and metabolic waste away from the fetus, while the placenta exchanges carbon dioxide and waste for oxygen and nutrients before oxygenated blood returns through the umbilical vein. During development, these vessels arise from the internal iliac arteries and provide a route for placental circulation. Their anatomy and blood-flow patterns support studies of fetal development, placental function, and prenatal health, including Doppler assessment of vascular resistance and fetal well-being.

This video protocol illustrates the isolation and culture of human umbilical vein endothelial cells (HUVEC) from human umbilical cord. Once isolated these cells can be used for in vitro angiogenesis assays like the Optimized Fibrin Gel Bead Assay also demonstrated by the Hughes lab.

Isolation of Precursor B-cell Subsets from Umbilical Cord Blood

Here we describe a protocol for isolating subsets of precursor B-cells from umbilical cord blood. A sufficient quantity and quality of nucleic acids may be extracted from the cells and used in subsequent assays utilizing DNA or RNA.

Isolation of Human Umbilical Vein Endothelial Cells and Their Use in the Study of Neutrophil Transmigration Under Flow Conditions

This article first describes a procedure for isolating human endothelial cells from umbilical veins and then shows how to use these cells to examine neutrophil transmigration under flow conditions. By using a low-volume flow chamber made from a polymer with the optical characteristics of glass, live-cell fluorescent imaging of rare cell populations is also possible.

The goal of this protocol is to isolate endothelial progenitor cells from umbilical cord blood. Some of the applications include using these cells as a biomarker for identifying patients with cardiovascular risk, treating ischemic diseases, and creating tissue-engineered vascular and heart valve constructs.
