Colonic Crypt Isolation

Colonic crypt isolation is a tissue-processing technique that separates the glandular units lining the colon from surrounding tissue for direct study. The method typically uses chelation to disrupt calcium-dependent cell adhesion, followed by gentle mechanical agitation and filtration to release and enrich intact crypts while limiting tissue damage. Isolated crypts retain epithelial organization and can be examined for morphology, gene expression, stem cell activity, and responses to experimental treatments. In cancer research, this approach supports comparisons between healthy and tumor-associated tissue, investigation of early epithelial changes, and development of ex vivo models for studying colorectal cancer biology and therapeutic responses.

Generation of Murine Primary Colon Epithelial Monolayers from Intestinal Crypts

In this protocol, we describe how to generate murine primary epithelial colon monolayers directly from intestinal crypts. We provide experimental approaches to generate confluent monolayers on permeable filters, confluent monolayers for scratch wound healing and biochemical studies, and sparse and confluent monolayers for immunofluorescence analysis.

We describe a technique that allows for the isolation of primary myofibroblasts from both mouse and human colon tissue, which can be utilized for in vitro experimentation.
