Epigenome-wide Screening

Epigenome-wide screening is the systematic analysis of epigenetic changes across the genome, helping researchers identify regulatory alterations that influence cancer development and progression. It typically compares tumor and normal samples using high-throughput assays that measure DNA methylation, histone modifications, or chromatin accessibility, then detects regions with significant differences in epigenetic state. In cancer research, these profiles can reveal biomarkers for early detection, tumor classification, prognosis, and treatment response. They also highlight disrupted regulatory pathways and may identify targets for epigenetic therapies, supporting more precise approaches to cancer biology and patient care.

2026

Here, we present a protocol for generating chemoresistant cells using a dose-escalation approach, followed by a CRISPR/Cas9-based screen using a focused sgRNA library targeting epigenetic modifiers to identify regulators of acquired chemoresistance. The protocol also provides multiple optimization points tailored to chemoresistant cell models, offering a robust framework for researchers investigating resistance mechanisms.
