Squalene-chidamide Prodrug

Squalene-chidamide prodrug is a molecularly engineered form of the histone deacetylase inhibitor chidamide that couples it to squalene, a lipid-like natural compound, to improve drug delivery in cancer research. The conjugate combines the pharmacological activity of chidamide with squalene’s hydrophobicity, enabling the amphiphilic molecule to self-assemble into nanoscale particles in aqueous environments and potentially release chidamide under biological conditions. This design can enhance formulation, cellular uptake, and tumor-directed delivery while reducing limitations associated with poorly water-soluble drugs. Squalene-chidamide systems therefore provide a platform for studying nanomedicine strategies that improve the therapeutic performance of epigenetic cancer treatments.

This protocol describes the fabrication and characterization of a photoresponsive prodrug-dye nanoassembly. The methodology for drug release from the nanoparticles by light-triggered disassembly, including the light irradiation setup, is explicitly described. The drugs released from the nanoparticles following light irradiation exhibited excellent anti-proliferation effects on human colorectal tumor cells.
