Her2 Targeting

HER2 targeting is a therapeutic and molecular-design strategy that selectively recognizes human epidermal growth factor receptor 2 (HER2), a cell-surface protein overexpressed in some cancers, to alter signaling or deliver treatment. Antibodies and other ligands bind the receptor’s extracellular domain, while small-molecule inhibitors can occupy its intracellular kinase site; antibody-drug conjugates additionally use a linker to transport a cytotoxic payload into HER2-positive cells. In chemistry, these approaches guide ligand optimization, conjugation chemistry, and assay development for selective cancer therapy, biomarker-based patient selection, imaging, and the design of more precise treatments.

Fluorescence in-situ hybridization (FISH) is often required in combination with histopathology and molecular diagnostics for selection of therapy in personalized medicine. A novel non-cross-linking, formalin-free tissue fixative that allows high quality morphologic, molecular and FISH analyses from the same specimen by addition of a post-fixation step before FISH is presented.

Heterogeneous distribution of HER2-positive cells can be observed in a subset of breast cancers and generates clinical dilemmas. Here, we introduce a reliable and cost-effective protocol to define, quantify, and compare HER2 intra-tumor genetic heterogeneity in a large series of heterogeneously processed breast cancers.
