Universal Donor Stem Cells

Universal donor stem cells are stem cells engineered or selected for compatibility with many recipients, addressing immune rejection that limits cell-based therapies. Typically, researchers use targeted gene editing to reduce or alter human leukocyte antigen display, which can lessen recognition by recipient T cells; successful designs must also avoid activating natural killer cells and other immune defenses. In developmental biology, these cells provide standardized material for studying differentiation, tissue formation, and disease mechanisms while supporting organoid research and regenerative medicine. Their potential to generate broadly usable cell therapies could simplify manufacturing, improve treatment access, and accelerate the translation of stem cell research.

Quantification of donor-derived cells is required to monitor engraftment after stem cell transplantation in patients with hemoglobinopathies. A combination of flow cytometry-based cell sorting, colony formation assay, and subsequent analysis of short tandem repeats may be used to assess the proliferation and differentiation of progenitors in the erythroid compartment.

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.
