Airway Epithelial Model

An airway epithelial model is a laboratory system that reproduces key features of the respiratory tract lining to study its barrier, immune, and host defense functions. Often established by culturing airway cells at an air-liquid interface, the model supports differentiation into basal, ciliated, and mucus-producing cells, creating tissue-like organization that can be exposed to microbes or inflammatory stimuli. In immunology and infection research, these models help investigators examine pathogen attachment and replication, epithelial signaling, mucus and ciliary clearance, and interactions with immune cells. They also provide a controlled platform for evaluating disease mechanisms, antiviral responses, and potential therapeutics.

Cell membrane–shed microparticles (MPs) are active biological vesicles that can be isolated and their pathophysiological effects investigated in various models. Here we describe a method for generating MPs derived from T lymphocytes (LMPs) and for demonstrating their proapoptotic effect on airway epithelial cells.

This paper describes different methods of growing Pseudomonas aeruginosa biofilms on cultured human airway epithelial cells. These protocols can be adapted to study different aspects of biofilm formation, including visualization of the biofilm, staining of the biofilm, measuring the colony forming units (CFU) of the biofilm, and studying biofilm cytotoxicity.
