Hypoxic Ischemia

Hypoxic ischemia is a pathological state in which tissue receives too little oxygen because blood flow is reduced or interrupted, making it a critical concern in medicine. Oxygen deprivation limits mitochondrial ATP production, disrupting ion balance and triggering excitotoxicity, oxidative stress, cellular swelling, and inflammation; restoring circulation can also cause reperfusion injury. These mechanisms are central to conditions such as ischemic stroke, cardiac arrest, and neonatal hypoxic-ischemic encephalopathy, where the timing and severity of injury influence neurological and systemic outcomes. Understanding hypoxic ischemia supports diagnosis, prognostic assessment, and development of strategies that preserve tissue during oxygen loss and reperfusion.

We performed unilateral carotid artery occlusion on postnatal day 7-10 CD-1 mouse pups to create a neonatal hypoxic-ischemic (HI) model and investigated the effects of HI brain injury. We studied neurobehavioral functions in these mice compared to non-operated normal mice.

A protocol is described that uses anoxia/starvation in C. elegans to model ischemia/reperfusion. Functional outcomes include increased mortality, visible abnormalities in GFP-labeled neuronal processes, and impaired behavioral responses that require neuronal function.

Thromboembolic stroke models are vital tools for optimizing the recanalization therapy. Here we report a murine thrombotic stroke model based on transient cerebral hypoxic-ischemic (tHI) insult, which triggers thrombosis and infarction, and responds favorably to tissue plasminogen activator (tPA)-mediated fibrinolysis in a therapeutic window similar to those in stroke patients.
