Platelet Concentrate Preparation

Platelet concentrate preparation is a transfusion-medicine technique that separates and collects platelets from whole blood or apheresis donations to support hemostasis in patients with thrombocytopenia or impaired platelet function. The process uses anticoagulated blood and controlled centrifugation or automated cell separation to isolate the platelet-rich fraction, followed by storage under conditions that preserve platelet viability and function. Prepared concentrates are administered to reduce bleeding risk before procedures or to treat active bleeding, while quality control evaluates platelet content, sterility, storage conditions, and clinical suitability. Standardized preparation improves transfusion safety and supports effective patient-specific care.

This article describes the process used by Örebro University Hospital to produce double dose buffy coat platelet concentrates prepared from whole blood donations and treated with the INTERCEPT Blood System for pathogen inactivation. The in vitro quality of the final platelet units are evaluated over 7 days of storage.

We describe a straightforward method for the isolation of washed platelets from human blood followed by agonist-induced platelet aggregation measurements by turbidimetry. As an example we apply this method for studying the aggregation response of human platelets to collagen after a pre-incubation with the Pannexin1 channel inhibitor Brilliant Blue FCF.

This paper describes an experimental model of hemostasis that simultaneously measures platelet function and coagulation. Platelet and fibrin fluorescence is measured in real-time, and platelet adhesion rate, coagulation rate, and onset of coagulation are determined. The model is used to determine platelet procoagulant properties under flow in concentrates for transfusion.
