Brainstem Isolation

Brainstem isolation is a neuroanatomical dissection technique that separates the brainstem from surrounding brain tissue while preserving its structural organization for focused study. The procedure uses recognizable landmarks and carefully placed cuts at defined anatomical boundaries to expose and remove the brainstem, often before sectioning, fixation, or preparation for physiological recording. Isolated brainstem tissue supports research on cranial nerve nuclei, respiratory and cardiovascular control, sensory and motor pathways, and neural circuitry. By reducing surrounding tissue, the technique enables targeted histological, molecular, electrophysiological, and pharmacological analyses of essential brainstem functions and their disruption in neurological disease.

The central respiratory drive is located in the brainstem. Spontaneous respiratory motor output from an isolated brainstem-spinal cord is recorded by placing an electrode on the fourth ventral root. This experimental approach is valuable for pharmacological investigations or the assessment of respiratory challenges and genetic manipulations on rhythmic motor behavior.

Auditory brainstem neurons of avians and mammals are specialized for fast neural encoding, a fundamental process for normal hearing functions. These neurons arise from distinct precursors of embryonic hindbrain. We present techniques utilizing electroporation to express genes in the hindbrain of chicken embryos to study gene function during auditory development.
