Endothelial Cell Stimulation

Endothelial cell stimulation is the activation of blood vessel lining cells by biochemical, mechanical, or environmental cues, a process that regulates vascular behavior and tissue communication. These signals engage endothelial receptors and intracellular pathways, altering cell shape, permeability, adhesion, migration, and the release of signaling molecules; in neuroscience, neural activity, inflammatory mediators, and changes in blood flow can influence these responses. Studying endothelial cell stimulation helps clarify neurovascular interactions, including how vessels support brain metabolism, respond to injury, and contribute to the blood-brain barrier. This knowledge informs research on cerebrovascular disease, neuroinflammation, and strategies for modifying vascular function.

Here, we describe a method to measure intercellular Ca2+-waves in cell monolayers in response to a local single-cell mechanical stimulus and its application to investigate the properties and regulation of gap junction channels and hemichannels.

We provide a method for isolating and culturing pure populations of heart valve endothelial cells (VEC). VEC can be isolated from either side of the cusp or leaflet and immediately following, underlying interstitial cell (VIC) isolation is straightforward.

Hematopoietic stem and progenitor cells (HSPC) derive from specialized (hemogenic) endothelial cells during development, yet little is known about the process by which some endothelial cells specify to become blood forming. We demonstrate a flow-cytometry based method allowing simultaneous isolation of hemogenic endothelial cells and HSPC from murine embryonic tissues.
