Glutathione Reduction

Glutathione reduction is the biochemical process that regenerates reduced glutathione (GSH), a major intracellular antioxidant, from oxidized glutathione (GSSG), helping maintain cellular redox balance. Glutathione reductase catalyzes this reaction by using electrons from NADPH to convert the disulfide bond in GSSG into two GSH molecules, allowing glutathione to neutralize reactive oxygen species and participate in detoxification. In neuroscience, this pathway supports antioxidant defenses in neurons and glial cells, where high metabolic activity and oxygen demand can increase oxidative stress. Measuring glutathione reduction can help assess redox homeostasis, mitochondrial or cellular dysfunction, and mechanisms relevant to neurodegenerative disease.

Glutathione S-transferases (GSTs) are detoxification enzymes involved in the metabolism of numerous chemotherapeutic drugs. Overexpression of GSTs is correlated with cancer chemotherapy resistance. One way to counter this phenotype is to use inhibitors. This protocol describes a method using a spectrophotometric assay to screen for potential GST inhibitors.
