Orco Marker Gene

Orco marker gene refers to the gene encoding odorant receptor co-receptor, a highly conserved component of insect olfactory receptor neurons. Orco proteins pair with odorant-specific receptors to form ligand-gated ion channels in neuronal membranes; odorant binding changes channel activity, allowing ions to enter and generating electrical signals that encode smells. Because Orco is expressed broadly across many olfactory receptor neurons, researchers use it to identify, label, and study these cells in the nervous system. Orco markers support analyses of sensory neuron development, olfactory circuit organization, odor-guided behavior, and genetic manipulation of insect smell pathways.

An example of adenoviral gene therapy in the human diabetic organ-cultured corneas is presented towards the normalization of delayed wound healing and markedly reduced epithelial stem cell marker expression in these corneas. It also describes the optimization of this process in stem cell-enriched limbal epithelial cultures.

The ability to produce transgenes for Caenorhabditis elegans using genomic DNA carried by fosmids is particularly attractive as all of the native regulatory elements are retained. Described is a simple and robust procedure for the production of transgenes via recombineering with the galK selectable marker.

Recombineering and Gene Targeting

2023

One of the most widely used tools in modern biology is molecular cloning with restriction enzymes, which create compatible ends between DNA fragments that allow them to be joined together. However, this technique has certain restrictions that limit its applicability for large or complex DNA construct generation. A newer technique that addresses some of these shortcomings is recombineering, which modifies DNA using homologous recombination (HR), the exchange between different DNA molecules based...
