B.f. Skinner

B. F. Skinner was an American psychologist and a leading figure in behaviorism, known for explaining how environmental consequences shape behavior. His work centered on operant conditioning, in which behaviors become more or less likely depending on reinforcement or punishment; experiments using the “Skinner box” measured how animals responded to controlled stimuli and schedules of reward. Skinner’s research influenced learning theory, classroom instruction, behavior therapy, organizational practices, and animal training. It also established experimentally grounded methods for analyzing observable behavior, while prompting lasting debate about the roles of cognition, free will, and internal mental processes in psychology.

A technique combining in situ tetramer staining and in situ hybridization (ISTH) enables visualization, mapping and analysis of the spatial proximity of virus-specific CD8+ T cells to their virus-infected targets, and determination of the quantitative relationships between these immune effectors and targets to infection outcomes.

Here, we describe a method that combines in situ MHC-tetramer staining with immunohistochemistry to determine localization, phenotype, and quantity of antigen-specific T cells in tissues. This protocol is used to determine the spatial and phenotypic characteristics of antigen-specific CD8 T cells relative to other cell type and structures in tissues.

Nanoparticles of indinavir, ritonavir, efavirenz and atazanavir were manufactured using wet milling, homogenization and ultrasonication. These nanoformulations, collectively termed nanoformulated antiretroviral therapy (nanoART), assessed macrophage-based drug delivery. Monocyte-derived macrophage nanoART uptake, retention and sustained release were determined. These preliminary studies suggest the potential of nanoART for clinical use.
