Headspace Collection

Headspace collection is a sample-preparation technique that isolates volatile compounds from a liquid or solid matrix by sampling the gas phase above it. In a sealed vial, analytes partition between the sample and its headspace until equilibrium is established, with distribution influenced by volatility, temperature, matrix composition, and partition coefficients; the vapor is then withdrawn for chromatographic analysis, commonly gas chromatography. In biochemistry, this approach supports measurement of volatile metabolites, fermentation products, solvents, and other compounds that may be difficult to analyze directly in complex biological samples. It reduces transfer of nonvolatile matrix components, protects analytical systems, and enables reproducible qualitative and quantitative measurements.
