Sandwich Assay Format

Sandwich assay format is an immunoassay design that detects and quantifies a target analyte by binding it between two complementary recognition molecules, providing high specificity for bioengineering measurements. In a typical workflow, an immobilized capture antibody binds the analyte, a second detection antibody attaches to another site, and a label generates a signal proportional to the analyte concentration after unbound components are removed. This format supports sensitive analysis of proteins, hormones, biomarkers, and other biological molecules in complex samples, making it valuable for diagnostics, biosensor development, bioprocess monitoring, and research on engineered biological systems.

The following protocol describes the procedure to assemble sandwich-like cultures to be used as an intermediate stage between bi-dimensional (2D) and three-dimensional (3D) cellular environments. The engineered systems can have applications in microscopy, biomechanics, biochemistry and cell biology assays.

Microtiter Dish Biofilm Formation Assay

The assay describes a rapid means to measure early biofilm formation in bacteria and fungi. This method uses a microtiter plate as the substratum for microbial biofilm formation, and the biofilm is visualized using crystal violet strain. The assay provides either a qualitative or quantitative assay for early biofilm formation.

Snap Chip for Cross-reactivity-free and Spotter-free Multiplexed Sandwich Immunoassays

We demonstrate a snap chip technology for performing cross-reactivity-free multiplexed sandwich immunoassays by simply snapping two slides. A snap apparatus is used for reliably transferring reagents from microarray-to-microarray. The snap chip can be used for any biochemical reactions requiring colocalization of different reagents without cross-contamination.

Three-Dimensional Culture Assay to Explore Cancer Cell Invasiveness and Satellite Tumor Formation

Cancer cells are embedded in a collagen gel and then sandwiched in an acellular fibrin gel to generate a 3D culture system in which the invasiveness and formation of satellite tumors may be monitored.
