Bacterial Identification

Bacterial identification is the process of determining the identity and characteristics of bacteria, an essential foundation for studying microbial diversity, disease, and environmental interactions. It combines observations of colony morphology and cellular features with differential staining, culture-based tests, biochemical reactions, and molecular methods such as nucleic acid amplification or sequencing. In biology, these approaches help distinguish closely related organisms, link bacteria to infections or ecological functions, and guide research on physiology and evolution. Accurate identification also supports antimicrobial susceptibility testing, outbreak investigation, food and water safety monitoring, and the development of targeted strategies for controlling or using bacterial species.

Bacterial Detection & Identification Using Electrochemical Sensors

We describe an electrochemical sensor assay method for rapid bacterial detection and identification. The assay involves a sensor array functionalized with DNA oligonucleotide capture probes for ribosomal RNA (rRNA) species-specific sequences. Sandwich hybridization of target rRNA with the capture probe and a horseradish peroxidase-linked DNA oligonucleotide detector probe produces a measurable amperometric current.

Matrix-assisted laser desorption/ionization time of flight mass spectrometry (MALDI-TOF MS) and molecular techniques (16S rRNA gene sequencing) permit the identification of rare bacterial pathogens in routine diagnostics. The goal of this protocol lies in the combination of both techniques which leads to more accurate and reliable data.

In this video, we describe a procedure for the expression of bacterial type III effectors in yeast and the identification of effector-induced growth inhibition phenotypes. Such phenotypes can be subsequently exploited to elucidate effector functions and targets.

Isolation and Identification of Bacterial Strains from Skin of Terrestrial Amphibians

This is a method for sampling and isolating culturable bacteria from the cutaneous microbiota of European plethodontid salamanders (Speleomantes genus). Here, we present a protocol for sampling salamander skin with swabs and processing them using culturable approaches. We describe sampling, isolation, and establishment of axenic culture with bacterial strain characterization.
