Gamma H2ax

Gamma H2AX, also written γH2AX, is the phosphorylated form of the histone variant H2AX and a widely used biomarker of DNA double-strand breaks. Following DNA damage, ATM and related protein kinases phosphorylate H2AX at serine 139 near the break, creating γH2AX that spreads across nearby chromatin and forms visible nuclear foci. Detecting these foci by immunofluorescence or other antibody-based methods enables researchers to measure DNA damage and monitor repair kinetics in cells. Gamma H2AX analysis supports studies of genotoxicity, radiation biology, cancer biology, and the effects of DNA-damaging drugs, although signal interpretation requires consideration of damage type and experimental timing.

The pedunculopontine nucleus (PPN) is located in the brainstem and its neurons are maximally activated during waking and rapid eye movement (REM) sleep brain states. This work describes the experimental approach to record in vitro gamma band subthreshold membrane oscillation in PPN neurons.
