Genetic Requirements Analysis

Genetic Requirements Analysis is a systematic method for determining which genes are necessary for a biological process, phenotype, or condition. It typically uses targeted or large-scale gene perturbations, such as knockouts, knockdowns, or inhibition, followed by comparison of growth, survival, development, or another measurable trait against appropriate controls. By linking gene disruption to changes in phenotype, researchers can distinguish essential genes from those that are dispensable, conditionally required, or functionally redundant. In biology, this approach supports pathway mapping, functional annotation, studies of host–microbe interactions, disease mechanism research, and identification of potential therapeutic targets.

This article describes a yeast growth-based assay for the determination of genetic requirements for protein degradation. It also demonstrates a method for rapid extraction of yeast proteins, suitable for western blotting to biochemically confirm degradation requirements. These techniques can be adapted to monitor degradation of a variety of proteins.
