Tetramer Production

Tetramer production is the laboratory generation of molecular complexes containing four copies of a defined component, a strategy that increases binding strength and enables precise detection of biological targets. In immunology, recombinant peptide–MHC molecules are commonly produced, purified, biotinylated, and assembled with streptavidin to create multivalent complexes that bind antigen-specific T-cell receptors. These tetramers can be fluorescently labeled and analyzed by flow cytometry to identify, quantify, and characterize immune-cell populations. The approach supports studies of adaptive immunity, vaccine responses, infection, cancer, and autoimmune disease, while providing a sensitive tool for linking antigen recognition to cellular function.

This procedure demonstrates the purification and in vitro expansion of antigen specific CD4+ T cells from whole peripheral blood and their visualization using MHC class II tetramers. Tetramers permit the direct visualization of T cells with a single antigen specificity and defined MHC class II restriction.

A Peptide-MHC-I Tetramer Staining Technique to Analyze SIV-Specific CD8+ Memory T Cells

2025

This video demonstrates a method for characterizing CD8+ memory T cells specific to simian immunodeficiency virus (SIV) using peptide-major histocompatibility complex class I (pMHC-I) tetramers. A pMHC-I tetramer is a complex of four MHC-I molecules bound to SIV-specific peptides and conjugated to a fluorophore. These tetramers selectively bind to T cell receptors (TCRs) on SIV-specific CD8+ T cells, facilitating the characterization of these cells.

Here, we describe a method that combines in situ MHC-tetramer staining with immunohistochemistry to determine localization, phenotype, and quantity of antigen-specific T cells in tissues. This protocol is used to determine the spatial and phenotypic characteristics of antigen-specific CD8 T cells relative to other cell type and structures in tissues.
