Kpc Mouse Model

The KPC mouse model is a genetically engineered model of pancreatic ductal adenocarcinoma that reproduces key features of human pancreatic cancer, including tumor development, stromal remodeling, and metastatic progression. In this system, Pdx1-Cre activates oncogenic KrasG12D and mutant Trp53R172H alleles in pancreatic progenitor cells, driving stepwise transformation from precursor lesions to invasive tumors. Researchers use KPC mice to study pancreatic tumor initiation, interactions between cancer cells and the tumor microenvironment, immune suppression, and treatment resistance. Because tumors arise in their native tissue and progress over time, the model supports evaluation of drug candidates, combination therapies, biomarkers, and mechanisms underlying poor clinical outcomes.

Multianimal Magnetic Resonance Imaging for Tumor Measurements in Pancreatic Cancer Mouse Models

This protocol uses a multichamber bed for parallel magnetic resonance imaging (MRI) of up to four animals to detect pancreatic adenocarcinoma tumors in genetically engineered mouse models. This multianimal MRI protocol is fast and cost-effective for detecting and measuring tumors, facilitating animal selection for preclinical studies and longitudinal monitoring of tumor growth.

Mouse Models of Periventricular Leukomalacia

We established mouse models of periventricular leukomalacia (PVL), the predominant brain injury in premature infants characterized by periventricular white matter lesions. Hypoxia/ischemia with/without systemic infection are the primary causes of PVL. Unilateral carotid ligation and hypoxia exposure with/without lipopolysaccharide injection creates PVL-like lesions in P6 mice.

We describe protocols for our mouse graft arteriosclerois (GA) models which involve interposition of a mouse vessel segment into a recipient of the same inbred strain. By backcrossing additional genetic changes into the vessel donor, the model can assess the effect of specific genes on GA.
