Rat Kidney Perfusion

Rat kidney perfusion is an ex vivo technique that maintains an isolated rat kidney by supplying it with a controlled perfusate, allowing renal function and tissue responses to be studied outside the animal. By cannulating the renal vasculature and circulating an oxygenated, nutrient-containing solution under regulated pressure and flow, researchers preserve aspects of filtration, vascular transport, and cellular signaling for a defined experimental period. In cancer research, this model can help evaluate how anticancer compounds, tumor-associated signals, or altered vascular conditions affect kidney tissue and drug delivery. Its controlled environment supports mechanistic studies, renal toxicity assessment, and therapeutic development.

The severe organ shortage has resulted in increased use of marginal kidney grafts for transplantation. This has triggered interest in alternative storage methods, since marginal grafts especially tolerate cold storage poorly. The technique of normothermic ex vivo kidney perfusion (NEVKP) represents a novel preservation method for kidney grafts prior to transplantation.

The administration of drugs for recovery of kidney function requires control of the localization and distribution of the therapeutic compound. Here, we describe in detail a simple technique for intrarenal delivery of drugs in rats. This procedure may be easily performed with no mortality and high reproducibility.
