L929 Cells

L929 cells are an established mouse fibroblast cell line used as a standardized in vitro model for studying cellular responses to chemicals, materials, and biological stimuli. In culture, these adherent cells proliferate under controlled conditions, and exposure to a test substance can change metabolic activity, membrane integrity, or growth, allowing viability to be quantified with assays such as MTT reduction. In chemistry and materials research, L929 cells support preliminary cytotoxicity screening, evaluation of extractables and biomaterials, and comparison of formulation or surface effects. Their reproducible growth and sensitivity to toxic insults make them useful for linking chemical composition with biological compatibility before more complex testing.

Here we describe a facile preparation of chitosan-based injectable hydrogels using dynamic imine chemistry. Methods to adjust the hydrogel’s mechanical strength and its application in 3D cell culture are presented.

Derivation of Hematopoietic Stem Cells from Murine Embryonic Stem Cells

This protocol details the derivation of transplantable hematopoietic stem cells from mouse embryonic stem cells (ESC) and their subsequent injection into lethally irradiated recipient mice. Briefly, ESC are differentiated as embryoid bodies, which are then infected with retroviral HoxB4 and co-cultured with OP9 stromal cells and hematopoietic cytokines.

Metaphase to anaphase transition is triggered through anaphase-promoting complex (APC/C)-dependent ubiquitination and subsequent destruction of cyclin B. Here, we established a system which, following pulse-chase labeling, allows monitoring cyclin B proteolysis in entire cell populations and facilitates the detection of interference by the mitotic checkpoint.
