Benign Strictures

Benign strictures are noncancerous, abnormal narrowings of a hollow organ or duct that restrict the passage of materials such as food, fluids, or bile. They typically develop when inflammation, injury, infection, surgery, or chronic irritation triggers healing with fibrosis, causing scar tissue to contract and reduce the lumen. In clinical practice, symptoms and severity depend on the affected site and may include difficulty swallowing, abdominal pain, vomiting, or impaired drainage. Evaluation commonly combines patient history, imaging, and endoscopy, while treatment may involve dilation, stent placement, medication, or management of the underlying cause to restore function and prevent recurrence.

In the present protocol, a urethral stricture induction was developed in Wistar rats, followed by urethral reconstruction with a buccal mucosa graft. A retrograde urethrogram and laser Doppler assessment were performed, validating urethral reconstruction (after stricture formation) and graft placement.

We describe the manufacture of compressed hormone pellets, as well as subcutaneous surgical implantation into mice. This strategy can be combined with the growth of cell and tissue xenografts under the renal capsule of athymic nude mice to evaluate hormonal carcinogenesis and regulation of benign prostate growth.
