Germ Cell Cysts

Germ cell cysts are organized groups of developing germ cells that remain connected after cell division, making them important models for studying gametogenesis and tissue development. They form through successive mitotic or meiotic divisions with incomplete cytokinesis, which leaves intercellular bridges linking sibling cells and permits coordinated growth, signaling, and distribution of cellular materials. In developmental biology, researchers use germ cell cysts to examine germline differentiation, oocyte and sperm formation, and the cellular mechanisms that establish reproductive tissues. Studies of cyst architecture also reveal how cell polarity, cell fate, and communication between germ cells shape fertility and reproductive development.

Germ Cell Transplantation and Testis Tissue Xenografting in Mice

Protocols for germ cell transplantation and testis tissue xenografting are described. Germ cell transplantation results in donor-derived spermatogenesis in recipient testes and represents a functional reconstitution assay for identification of spermatogonial stem cells (SSCs). Testis tissue xenografting reproduces testis development and spermatogenesis of various donor species in recipient mice.

Genes essential for survival pose technical hurdles for creating mutant lines. Leapfrogging circumvents lethality by combining genome editing with primordial germ cell transplantation to create wild-type animals carrying germline mutations. Leapfrogging also permits the efficient generation of homozygous null mutants in the F1 generation. Here, the transplantation step is demonstrated.
