Pma Differentiation

PMA differentiation is a cell-culture method that uses phorbol 12-myristate 13-acetate (PMA) to induce immature cells, commonly monocytic cell lines, to acquire a more specialized phenotype for developmental biology studies. PMA activates protein kinase C signaling, triggering changes in gene expression, cell morphology, adhesion, and immune-cell function that produce macrophage-like characteristics in responsive cells. Researchers use this approach to model stages of hematopoietic differentiation, investigate cellular signaling and immune development, and generate standardized cell populations for experiments. Because differentiation depends on exposure conditions and cell type, careful control of PMA concentration and treatment duration is essential for reproducible outcomes.

This protocol is based on LPS and ATP-induced death of PMA-differentiated THP-1 macrophages. We use flow cytometry to analyze Annexin V and 7-AAD double staining to detect cell death, using the whole cell and employing scanning electron microscopy to observe the cell membrane morphology.

This video demonstrates the in vitro differentiation of granulocyte-macrophage-colony-stimulating factor or GM-CSF-producing T helper cells (THGM).
